How to choose a right capillary column?
1. Column length
The resolution is proportional to the square root of the column length. Under other conditions unchanged, 4 times the column length is required to obtain doubled resolution. Shorter columns are suitable for simpler samples, especially those composed of components that differ greatly in structure, polarity, and volatility.
15m short column is used to quickly separate simpler samples and is also suitable for scanning analysis;
30m chromatographic column is the commonly used column length, most of the analysis is done on this length of the column;
50m/60m or longer chromatographic columns are used to separate more complex samples.
2. Column inner diameter
The inner diameter of the column directly affects the efficiency, retention characteristics and sample capacity of the column. Small-bore columns have higher efficiency than large-bore columns, but the column capacity is smaller
0.25mm: Has higher column efficiency and lower column capacity. It is better to separate complex samples.
0.32mm: The column efficiency is slightly lower than 0.25mm, but the column capacity is about 60% higher.
0.53mm: It has a column capacity similar to a packed column and can be used for split injection or splitless injection.
When performing trace analysis, a large-bore capillary column is more appropriate.
3. Liquid film thickness
The thickness of the liquid film affects the retention characteristics and capacity of the column. As the thickness increases, the retention time also increases.
0.1-0.2um: The thin liquid film capillary column elutes components faster than the thick liquid film capillary column, the required column temperature is lower, and the column loss is smaller at high temperature, which is suitable for the analysis of high boiling point compounds.
0.25-0.5um: commonly used liquid film thickness.
0.6-5.0urn: thick liquid film, which is more advantageous for the analysis of low boiling point compounds.
