Real-Time PCR for avian influenza virus (aiv) Rapid Detection Kit

Description

Avian Influenza Virus (AIV) RT-qPCR test kit enables the safe detection of Avian Influenza Virus RNA in oropharyngeal/tracheal swab,trachea,lung,spleen,large intestine from poultry (chicken/turkey) within 100 minutes. The kit consists of fluorescence PCR mix , enzyme mix , positive control and negative control for RT-qPCR amplification of AIV. Fluorescence PCR mix contains primers, probes and PCR buffer for detection of Avian Influenza Virus.

Operation:

1. Sample preparation
(1) Swabs
a. Place one oropharyngeal/tracheal swab sample into 2 mL centrifuge tube, then add 1 mL sterile saline. Vortex vigorously for 1 minutes. Centrifugation for 2 minutes with 8000×g, take out the supernatant.
b. Follow the commercial RNA extraction kit manual for the RNA extraction of
samples,positive control and negative control.

(2) Organs
a. Take 0.1 g sample(about the size of a soybean) to 2 mL centrifuge tube, then add 1 mL sterile saline, grinding and mixing. Centrifugation for 2 minutes with 8000×g, take out the supernatant.
b. Follow the commercial RNA extraction kit manual for the RNA extraction of samples,positive control and negative control.

2. PCR
(1) Take out the reagents in the room temperature for melting, mixing and vortex.
Centrifuge briefly.

(2) Prepare the RT-PCR master mix : combine the following components for the
number of reactions required plus 10% overage:

(3) Dispense 20 μL of RT-PCR master mix to the appropriate number of PCR strip tubes.
(4) Add 5 μL of sample supernatant to each well (include the positive control and negative control for each test). The final reaction volume should be 25 μL.
(5) Seal each PCR tube, then centrifuge briefly to bring the contents to the bottom of the plate tubes.
(6) Write down the order of each sample.
(7) Set up PCR amplification cycling program as following:
