Furacillin Metabolite Rapid Detection Kit
This rapid test is used for detection of furacillin metabolite in aquatic (fish, shrimp) tissues. It takes about 80 minutes for small batch（6 samples）to detection process. It is apply to all kinds of enterprises and testing institutions. The detection limit of this product for furacillin metabolite is 0.5 μg/kg (PPB).
The furacillin metabolite rapid detection card uses the principle of competitive suppression immunochromatography. The furacillin metabolite in the sample conbines with the specific anti-furacillin metabolite monoclonal antibody labelled with colloidal gold during the flow process, which inhibite the binding of the antibody to the furacillin metabolite -BSA conjugate on the test line of NC membrane.
If the content of furacillin metabolite in the sample is more than 0.5 μg/kg, the test line T is lighter than QC line C or the T line is no color, the test result is positive. On the contrary, the color of test line T is equal to or darker than that of quality control line C, and the test result is negative.
Specimen requirements
Fish, shrimp and other aquatic samples shall be collected in clean containers or self-sealed bags; If the sample cannot be sent for inspection in time, the fat should be removed, cut into pieces with scissors or ground with a meat grinder, and stored separately at -20℃. Repeated freeze-thaw is avoided.
Sample treatment
(1) Take a certain amount of samples (fish, shrimp, etc.) and cut them into pieces with scissors or grind them into pieces with a meat grinder after removing the fat;
(2) Take 5 g of treated samples to be measured in a 50 mL centrifuge tub. Add 5 mL Reagent A and 0.5 mL Reagent B in turn, shake and mix well for 3 minutes;
(3) Incubate for 10 minutes at 80℃ in water bath;
(4) Shake and mix well for 3 minutes after add 2 mL Reagent C and 8 mL Reagent D in turn;
(5) Take the supernatant 4 mL to 10 mL centrifuge tube after centrifuge for 5 minutes at 4000 RPM. Blow the solution dry with nitrogen or air at 65 ℃;
(6) Add 2 mL n-hexane, shake and mix well for 15 seconds after add 0.6 mL last sample solution accurately with a pipettor;
(7)Take the lower solution to be measured and remove the upper organic phase after centrifuge for 1 minutes at 4000 RPM (the centrifugation time can be extended to 5 minutes, if the delamination is not significant ).
Testing method and interpretation of results
(1)Return the unopened detection card and sample solution to room temperature before detecting;
(2)Take out the detection card and enzyme marker holes from the original aluminum foil bag and use it within half an hour;
(3)Use pipette to absorb 80 μL of sample solution into the enzyme-labeled well, blow repeatedly until the red substance in the well is completely dissolved, wait for 2 minutes;
(4）Lay the detection card flat, drop all the solution in the suction hole into the sample well, and start the timing after the sample is added;
(5)Judged according to the schematic diagram after 5 minutes for reaction, it will invalid in other time.
Negative results: T line is deeper than C line, or the same depth, indicating that the concentration of furacillin metabolite in the sample is below its detection limit;
Positive results: T line is shallower than C line, or T line is green, indicating that the concentration of furacillin metabolite in the sample is higher than its detection limit;
Ineffective reaction: if the test result C line does not appear, the reagent has failed.
